Intramolecular Excimer Fluorescence of Pyrene Maleimide-Labeled Dithiothreitol (Pyrene-DTT)

Abstract

The fluorescence properties of pyrene-DTT were studied as a model system for pyrene-tropomyosin for which excimer fluorescence has been reported (Betcher-Lange and Lehrer, 1978). DTT in dimethyl formamide or dimethyl sulfoxide was reacted at its 2 SH-groups with pyrene maleimide to form (S-[N-(l-pyrene)succinimido])2-DTT (pyrene1 -DTT). The product with cleaved succinimido-rings, pyreneII -DTT was also prepared by incubation of pyreneI -DTT in 1 mM NaOH. Pyrene-β-mercaptoethanol (βSH) adducts were similarly prepared. For spectroscopic measurements, 1 µM solutions were studied by dilution into appropriate solvents.
