Techniques for Ex-Vivo Bone Marrow Treatment with Immunotoxins

Abstract

The high in vitro potency of immunotoxins (ITs) demonstrated by several laboratories (Möller, 1982; Moolten et al, 1982; Neville and York, 1982; Raso, 1982; Thorpe and Ross, 1982; Vitetta et al, 1982; Jansen et al, 1982) is already used for ex-vivo bone marrow purging and especially for T-cell purging in allogeneic bone marrow transplantation (BMT) to prevent GvHD (Vallera et al, 1983; Jansen et al, 1985; Casellas et al, 1985). Since 1984 our group has treated about sixty patients with an anti-T A-chain IT. This experience allowed us to define the parameters needed to obtain sufficient T-cell cytoreduction with A-chain IT to prevent graft versus host disease (GvHD).
