The Role of Defective Interfering (DI) Particles in Viral Infection

A special class of viral deletion mutants was first detected biologically about 35 years ago and characterized physically about 20 years ago. They are formed by almost every virus group and given the name defective interfering (DI) particles. DI particles have the following properties:
  1. i)

    they are derived from infectious standard virus;

  2. ii)

    they contain the same structural proteins as standard virus;

  3. iii)

    they contain nucleic acid deletions and/or rearrangements;

  4. iv)

    they amplify themselves within the viral population, generally, but not necessarily always, at the expense of the standard virus (interference); and

  5. v)

    they require helper standard virus for their complete replication.
