Abstract
The term isoenzyme is applied to proteins that catalyze the same chemical reaction, yet are structurally different. This heterogeneity of enzymes has been known for several decades (Vessell, 1958; Cahn, 1962; Markert, 1963; Nisselbaum, 1963). In the last few years, isoenzyme determinations have become very important in the clinical laboratory because they have proven to be powerful diagnostic tools since the various tissues and organs of man exhibit characteristic isoenzyme profiles. When an infusion of particular isoenzymes into the circulation occurs due to damage to a specific tissue or organ, an isoenzyme determination can often pinpoint the site of injury. After a myocardial infarction (MI), isoenzymes of lactate dehydrogenase (LD) and creatine kinase (CK) are released from the myocardial tissue into the circulation. We have utilized the different immunochemical properties of CK and LD isoenzymes to develop simple, rapid and precise immunoassays for determining the specific cardiac isoenzymes of both enzymes.
