Abstract
To facilitate the genetic analysis and manipulation of the 3-chlorobenzoate degradative functions, it is of interest to clone DNA segments that carry individual genes and blocks of genes that encode parts of or complete catabolic pathways. The construction of “gene banks” will allow the isolation of hybrid plasmids that collectively contain all the endonuclease-generated fragments of the 3-chlorobenzoate degradative plasmid. The pMMB33 cosmid (13.75 kb) we use allows a high expression of the genes from P.sp.B13 in JE. coli and can be mobilized in Pseudomonas. This poster describes the cloning procedure and the analysis of gene expression.
