Specificity Criteria for Receptor Sites in Binding Studies

Abstract

A ‘binding site’ is not synonymous with ‘receptor site’: biochemical criteria [saturable, displaceable, high affinity, stereospecific,……) remain insufficient. There is no receptor without physiological response. Criteria are discussed with a number of examples, positive and negative. Also advocated is the postulate of receptor homogeneity, such that receptor sites fora given neurotransmitter are identical in all parts of the body: any odd results should not be regarded immediately as implying the occurrence of a receptor sub-type. Some reports of receptor sub-types, and even certain binding assays, are open to question. Aspects now considered include observations on 3H ligand trapping in intact cells, interpretation of drug displacement curves, correlation with pharmacological potency, regional and subcellular distribution of some brain receptors.
