Cytoprotection and Enteropooling by Prostaglandins

Abstract

Prostaglandins (PG) influence a variety of gastrointestinal functions. One of the most prominent effects is the inhibition of gastric secretion by several PG of the E and A type (1,2,3). Presumably because of this anti-secretory effect, these PG were also shown to inhibit ulcer formation (4,5,6) and to accelerate the rate of ulcer healing (7,8). More recently, another property of several PG, called “cytoprotection”, was reported. It is the ability to protect the mucosa of the stomach and the intestine against inflammation and necrosis when this mucosa is exposed to noxious agents. Such noxious agents include nonsteroidal anti-inflammatory compounds (NOSAC), (9,10,11) such as aspirin and indomethacin, necrotizing agents such as absolute ethanol, 0.6 N HC1, 0.2 NaOH, 80 mM taurocholate, and even boiling water (12,13,14,15,16,17). The severe gastric hemorrhagic lesions produced by any of these agents are completely prevented by a variety of PG. In this report, we will present evidence showing that cytoprotection is a property separate from the antisecretory effect of PG.
