On-line Determination of Reaction Rate Versus Substrate Concentration for an Enzymatically Catalysed Reaction by Means of a Microcomputer System
A microcomputer is used to record the change of conversion as a function of time during an enzymatically catalysed reaction. An electrical signal, which is proportional to substrate concentration is digitized and stored in constant time intervals. A sliding polynomial regression is used to smooth the concentration time curve and used to calculate the reaction rate from the first derivative of the determined polynoms (Figure 1).
