On Network Theory and H-2 Restriction

Abstract

The network hypothesis of Jerne (1974a,b, 1976) is based on the fact that the V (variable) regions of antibody molecules can function as antigens. Such antibodies can therefore stimulate and/or suppress responses of cells that have receptors which specifically recognize these V regions. One network model that incorporates this postulate is the “plus-minus network theory” (Hoffmann, 1975, 1978). In this paper a more extensive review of the data relevant to the theory is presented, and a number of phenomena are discussed within the framework of the theory that were not previously treated. These phenomena include the subclasses of T cells and H-2 restriction.
