In Vivo Activation of Murine Peritoneal Macrophages by Nocardia Water Soluble Mitogen (NWSM)

Abstract

The water soluble fraction extracted from various strains of Nocardia, and hereafter referred to as NWSM (12), is a potent B-cell mitogen in several strains of mice (7) including the C3H/He Orl. mice, which are low responders to lipopolysaccharide (LPS) (10), and also in rabbits (6) and in humans (9). Bacterial lipopolysaccharides are also B-cell mitogens and adjuvants (19,14) in mice in vivo. However, in contrast to LPS, when NWSM was administered to mice, it failed to induce in vivo proliferation of splenic lymphocytes, polyclonal activation and appearance of PFC directed against syngeneic, bromelain-treated erythrocytes and thymocytes (18). These findings strongly suggest that the adjuvant activity of NWSM is not necessarily related to in vivo mitogenicity, and that other cells than B lymphocytes may be its primary target cell in vivo. During investigations of NWSM in vivo effects we examined the influence of this adjuvant on murine peritoneal macrophages in comparison to other water soluble adjuvants.
