Studies on Antibody to Bradykinin

A number of investigators have reported the production of antibody to bradykinin in rabbits (1–5). These workers conjugated bradykinin with proteins or synthetic polypeptides since bradykinin by itself is nonimmunogenic. In our laboratory we have found that human LMW kininogen I, which is thought to contain the kinin moiety at the carboxyl terminus, is able to compete with bradykinin for its antibody. Further, immunization of one sheep with this kininogen elicited high-affinity antibody to bradykinin as well as precipitating antibodies to the remainder of the kininogen molecule. Attempts to repeat this result in another strain of sheep, the original strain being no longer obtainable, were unsuccessful. This prompted us to explore other, more promising immunogens. The brady-kinin-ovalbumin conjugate, first described by Talamo and coworkers (4), appears to be the best.
