Immunologic Restoration of X-Irradiated Spleen by Repopulation with Circulating Lymphocytes

In previous reports [1, 2, 3, 4] it has been shown that heavy local X-irradiation of the spleen, which in the rat is the main site of hemolysin synthesis, totally destroys the immunocompetent lymphoid tissue. The irradiated spleen, however, when antigen is injected intravenously just prior to irradiation, has rapid and complete restoration of its hemolysin-forming capability due to repopulation with immunocompetent cells. The reticuloendothelial cellular structures of the spleen, which survive irradiation, most probably play an essential role in this restoration because they serve as framework for repopulating lymphoid cells and because they store the injected antigen. Serologically, restoration is manifested by the appearance of a normal (i.e., optimal) hemolysin response that is only delayed for about 2 days. Histologic studies [2] indicate that the immunologic recovery is probably due to repopulation with circulating lymphocytes, but such evidence cannot be entirely conclusive.
