Studies on the Mechanism of Action of Choline Acetyltransferase

Choline acetyltransferase (CAT) is the enzyme responsible for the formation of the essential neurotransmitter ACh by catalyzing the alcoholysis of acetyl CoA (AcCoA). This reaction is favored only slightly thermodynamically under physiological conditions (19), probably because ACh itself is a high-energy compound (12). It is likely that this is the case because the coulombic interaction between the trimethylammonium group and the acyloxy oxygen of this ester (16) favors nucleophilic attack on the carbonyl carbon.
