T Cell Modification of B Cell Responses to Antigen in Mice

Abstract

The antigen-reactive cells1 required for expression of cell-mediated and humoral immunities have been designated T cells2 and B cells, respectively (Roitt et al, 1969). The circulating pool of lymphocytes (Gowans and McGregor, 1965) is a mixture of T and B cells in mice (Sprent and Miller, 1972) and rats (Howard et al, 1972; Strober, 1972), and, as a consequence, all peripheral lymphoid organs (e.g., spleen and lymph nodes), lymphatic channels such as the thoracic duct, “unorganized” lymphoid aggregates, and the blood stream must contain both cell types. Antigens presumably make contact with reactive cells in many if not all of these lymphoid sites, and popular belief is that B cells have the option of differentiating to antibody-secreting cells, such as plasma cells, whereas T cells do not. The spectrum of T cell immunological activities has been extended to include a regulatory role in humoral antibody production. T cells and B cells, both with specificity for antigenic determinants, collaborate in numerous antibody responses in vivo and in vitro. The purpose of this paper is to examine existing data on the antibody-producing activity of B cells responding to antigens under conditions of T cell insufficiency in mice. There are numerous reviews on this topic in the literature, and the approach here, at least initially, will be somewhat negative. This hopefully does not reflect my attitude to the problem; an attempt is made to simply highlight difficultiesAntigen-reactive cell-antigen-sensitive cell; immunologically competent cell of interpretation of several experimental findings as well as stating the positive conclusions which have been drawn.
