The Primary Structure of Individual Transfer RNAs
Research into the primary structure of tRNA essentially began in 1958 when Holley began his systematic study of the fractionation of soluble RNA from tissue homogenates by means of the countercurrent method. The first experiments were sufficient to demonstrate the promising nature of this method, for the results showed wide distribution of the uv-absorbing material with the differentiation of separate maxima of acceptor activity relative to the various amino acids (Holley and Merrill, 1959; Holley et al., 1959).
