Abstract
Phosphonoformic acid was synthesized In 1924 by Nylen (1) and has been investigated recently as an antiviral agent because of its inhibition of viral DNA polymerase (2,3,4). Herpes simplex virus polymerase activity is quite sensitive to PFA Inhibition (Ki=3.5 uM) (2). Partially purified mammalian (calf thymus) DNA polymerase is less sensitive to PFA Inhibition, and this sensitivity depends on the enzyme concentration (K.>3.5 uM) (2).
