Abstract

There is now widespread appreciation for the fact that overall and local changes in protein conformation are critical to the mechanism of enzymatic catalysis. Yet, it has been difficult to document the occurrence and judge the significance of such changes, since methods for the analysis of protein structure often require conditions which cannot be compared easily and directly with those designed for determining function. This is true even while the number, resolving power and sophistication of suitable approaches has increased rapidly.
