Abstract

Selective concepts of antibody formation imply that preformed antibodies are present as receptors on the membrane of immunocytes and are instrumental, after interaction with the corresponding antigens, in triggering these cells to undergo a series of events that ultimately lead to antibody production. This view of the physiology of the immune response was first put forward by Ehrlich (1900), who visualized polyvalent immunocytes, each carrying on its membrane a variety of receptors for antigens; the interaction of antigen with one kind of these receptors induced a “compensatory” hyperproduction of the receptors, which then spilled into the body fluids as soluble antibodies. Almost 60 years later Burnet (1959) proposed a similar concept, but with one major difference, namely, that the combining property of the receptors present on the membrane of one cell is uniform, and that the interaction of these receptors with the corresponding antigen determines this cell to clonal expansion or deletion with the corresponding consequences of antibody production or tolerance.
