Abstract

Oligodeoxyribonucleotides of defined sequence may now be synthesised as a matter of routine at a reasonable cost in both time and materials. As both applications of and synthetic methods for oligodeoxyribonucleotides are much more highly developed than those for oligoribonucleotides, the term ‘oligonucleotide’ has come to be synonymous with oligodeoxyribonucleotide and will be used in this way in this chapter. An oligonucleotide of 15–35 nucleotides may be synthesised and purified in 3–5 days using simple apparatus and well-established methodologies. Slightly more sophisticated approaches allow the ‘simultaneous’ synthesis of 2 to about 125 oligonucleotides in a similar time.
