Abstract

Sensitive methods for detecting antibody production by individual cells (in the primary response) have been developed, which maintain cell integrity and adequately measure antibody specificity. Among these methods are several plaque-forming procedures (1) (2) (3). While the methods are sensitive and specific, cytological identification of the cells has been hampered by the difficulty of their rescue from the medium. In this study, micromanipulation of cells from carboxymethyl cellulose (CMC) gels (4) has been followed by cytology and electron microscopy (to be reported at a later date). By this procedure, it has been possible to identify the cells involved in antibody production and to determine their frequency.
