Abstract
In this chapter, we summarize some current approaches to the de novo design of structurally defined polypeptides with emphasis on their application to synthetic peptide vaccine development and review our attempts at engineering conformational peptides for vaccines. The purpose of this review is twofold:

1.
to briefly enumerate strategies of synthetic peptide design as it pertains to elucidating and mapping B-cell antigenic determinants, and
-
2.
to view this field, and specifically synthetic vaccines from design principles and perspectives, as it relates to the topographic nature of antigenic sites recognized by B cells.
