Abstract

Since prostaglandins have been shown to play a regulatory role in a number of biological systems, it is not surprising that their role in oncology has been fairly carefully studied. The recent explosion of data on the relationship between prostaglandins and tumor cell growth and function has been extraordinary. Based on earlier in vitro observations, newer studies have implicated prostaglandins in abnormalities in humans. This chapter will attempt to review and interpret the pertinent observations, both in vitro and in vivo. Since the general interactions of prostaglandins with the adenylyl cyclase-cyclic AMP system have been extensively reviewed, they will be alluded to only briefly. Two major aspects of the interactions between prostaglandins and tumor cells will be emphasized, prostaglandin biosynthesis by tumor cells and its effect on replication and the bone-resorbing effects of prostaglandins and their role in the hypercalcemia of neoplastic diseases.
