Abstract
There are two general types of Chromatographic techniques, planar and column systems. In the first of these the separation is usually terminated after a fixed time and the different distances moved by each zone are measured. In column techniques the path length for each compound in a mixture is the same (i.e. the full length of the column) and it is the time taken to travel this distance (or the volume eluted under constant flow conditions) which is measured. However, it should be noted that when preparative chromatography is employed the elution of fixed-volume fractions in numerical sequence is most often used and little attention is paid to the actual time involved.
