Protein Conformation Affects the Efficacy of Pertussis Vaccines

Abstract

Production of new and better vaccines to improve man’s health is one of the goals of recombinant DNA technology. In spite of the great number of papers published on the cloning and expression of genes from a variety of human and animal pathogens, as yet only the hepatitis B recombinant vaccine has been successfully introduced for human immunization and there are not many other promising candidates foreseen in the near future. The general finding has been that recombinant antigens are immunogenic but induce antibodies which do not recognize, or recognize poorly, the antigens in their native conformation and therefore they are not effective vaccines. This finding is particularly true when the natural antigens have a complex structure as in the case of viral capsids or proteins composed of multiple subunits.
