Abstract
Biogenesis of the photosynthetic membranes of chloroplasts and cyanobacteria involves intricate interplay between many complex processes such as translocation of lumenal proteins across the membranes, integration of transmembrane and peripheral proteins in the lipid bilayer, their association with pigment and metal cofactors, and assembly of individual subunits into larger complexes. Assembly of photosystem I (PS I) involves most of these complexities, thus providing a model system to study biogenesis of protein complexes in the thylakoid membranes. PS I catalyzes the photooxidation of plastocyanin and photoreduction of ferredoxin (Bengis and Nelson, 1975). The PS I preparations isolated from thylakoid membranes of chloroplasts and cyanobacteria comprise of seven or more polypeptides (Fig. 1), approximately 100 chlorophyll a molecules, several beta-carotenes, a pair of vitamin K1 molecules and three iron-sulfur clusters (Chitnis and Nelson, 1991). PS I is localized predominantly in the nonappressed regions of thylakoids in chloroplasts of higher plants.
