Abstract

G protein -coupled receptors mediate the actions of a wide variety of extracellular signals, including numerous neurotransmitters and hormones,chemoattractants,cytokines and sensory stimuli such as photons and odorants,which regulate key physiological functions. A general property of these receptor systems is that their acute or sustained exposure to agonists often leads to a rapid loss of receptor responsiveness, a process termed desensitization,tachyphylaxis or tolerance. Such regulatory processes are essential for modulating the sensitivity of cells to messengers and allow the adaptation of cells to different signaling environments,thus contributing to signal integration and plasticity (Lefkowitz, 1993a; Vaello et al., 1994).
