Chemiluminescence and the Challenge to Attomole Detection

Abstract

Liquid chromatography (LC) has become a major tool for the determination of a wide variety of samples using refractive index, UV and fluorescence detectors. Unfortunately, the sensitivity and selectivity of these detection methods often do not meet the requirements of modern trace-level analysis. Fluorescence is frequently selected as the preferred method of detection if low concentration of analytes have to be determined. Since the range of compounds displaying strong native fluorescence is relatively small, derivatization and /or analyte conversion play an important role in LC with fluorescence detection. However, in many cases derivatization does not allow detection in the low or sub-nanogram/ml range which is increasingly demanded today.
