Early Events in the Generation of Antigen-Specific Cytotoxicity in MLC

Abstract

The generation of cytotoxicity in vitro has been shown to be accompanied by proliferation of the responding lymphocytes. Interference with the proliferative phase of the reaction has been shown to decrease the magnitude of the cytotoxic response as measured several days later (1–3); moreover, proliferation can occur in MLC without leading to cytotoxic effector cells (4,5) Two key points yet to be established, however, are whether or not one or more rounds of cell division are absolutely required for the initial expression of cytotoxicity in response to alloantigens, and whether or not appearance of cytotoxic function in responding cells requires expression of new genetic information
