Abstract
In response to Kupfer’s comment about the need for caution in the interpretation of assays for benzpyrene hydroxylase activity, Schenkman again emphasized that most benzpyrene hydroxylase assays monitor the appearance of a fluorescent metabolite, despite the lack of stoichiometric relationship between the fluorescent compounds produced and the disappearance of benzpyrene. It has been shown that fluorescent metabolites other than the 3-hydroxy derivative are produced by the AHH-mediated reaction, some of which have not been identified; thus their fluorescence coefficients are not known. Nonfluorescent conjugates are also produced from the hydroxylated intermediates in a microsomal system. Thus he agreed that great care must be exercised in interpreting all results obtained in fluorescent assays of benzpyrene hydroxylase activity.
