A Reexamination of Messenger RNA Populations in the Preimplantation Mouse Embryo

Abstract

Regulation of gene expression and the flow of genetic information from the nucleus to the cytoplasm in eukaryotic cells are controlled at a number of levels, beginning with the transcription of DNA to produce a primary RNA transcript. These transcripts are subjected to a number of posttranscriptional modifications that can include processing through splicing and ligation of different regions of the RNA molecule, capping and methylation of the 5′ end, and polyadenylation of the 3′ end of the RNA. Additional posttranscriptional controls related to association of mRNAs with ribonucleoproteins (mRNP) and translational controls at the level of binding of mRNA to ribosomes and subsequent steps in the synthesis of polypeptides also are involved. Similar types of controls can be expected to operate in differential gene expression during preimplantation development. The problem resides in assessing the extent to which the various controls are involved in regulating the genetic activity of different cells at various stages of the early developmental process.
