Abstract

Populations of polyadenylated messenger RNA (mRNA) sequences can conveniently be characterised by studying the kinetics of hybridisation between the mRNA population and a cDNA copy population which is usually synthesised by using reverse transcriptase.1,2 The most common method is to anneal unlabelled mRNA with radioactively labelled cDNA, and to measure how much of the latter has become duplexed at different times by treating successive samples of the annealing mixture with nuclease S1.
