Effect of Gossypol on Prostatic Androgen Receptors in Male Rats

Abstract

In China, gossypol has been given to more than 8000 human volunteers over a period of three years without any overt toxicity; the drug has been reported to be 98.4% effective [1]. Apart from some preliminary observations, the precise mechanism of action of gossypol is not clear. It has been reported that it inhibits testosterone production in the rats [2] and impairs accessory sex organs function in mice and rats [3, 4]. Chinese researchers have reported a decrease in the weight of the ventral prostate after gossypol treatment [6], and these same investigators have further observed that the weight of the ventral prostate can be restored to normal after testosterone administration [5]. The impairment of prostate function after gossypol treatment could be due to inhibition in testosterone release/synthesis or to the direct effect of gossypol on the prostate. Coulson and his associates have suggested that gossypol might act as an antiandrogen [6]. The present investigations have been designed to examine the direct effect of gossypol on prostate androgen receptors.
