Implications of Atrial Natriuretic Factor (ANF) in the Pathogenesis of Experimental and Human Hypertension

Abstract

It is now well established that the atria in mammals (1–3) and possibly the atria and ventricle in non-mammalian vertebrates (4,5) are an endocrine gland harbouring a peptide (atrial natriuretic factor, ANF) of nown composition whose cDNA and gene have been cloned (6,7). The main circulating form of ANF in the rat is the C-terminal, active portion of the molecule (Ser 99-Tyr 126) (8) while a minor circulating component is a truncated version (Ser 103-Tyr 126) (9). Secretory granules of rat atrial cardiocytes harbour the whole propeptide (Asn 1-Tyr 126) (10) and the pre portion of the peptide is made up of 24AA. The two Arg-Arg at the C-terminal end of the molecule which have been found by cloning of the cDNA and gene are not part of the circulating molecule in the rat and are not present in the human prepropeptide (1).
