The Cell Biology of Antigen Processing: A Review

Abstract

In the last 13 years the major mechanisms whereby the immune system generates an extremely large number of different antibodies and T cell antigen receptors were identified. By contrast, it is only in the last 4 years that any detailed understanding of a third aspect of antigen recognition has emerged; namely, how an antigen presenting cell creates a specific ligand for recognition by the T cell antigen receptor. Fortunately, the methods of molecular and cell biology, combined with current immunological paradigms, offer a unique opportunity to attack the mechanisms of antigen presentation. The following review will attempt to place antigen processing in the context of our current knowledge of the expression of the MHC molecules that serve as “receptors” for immunogenic peptides on the antigen presenting cell (APC) side of an immunologic cell couple.
