Abstract

Marker plasmids have been constructed to enable the detection and survival of populations of Gram-negative bacteria and to assess their ability to survive after release into lakewater (Winstanley et al., 1989; 1991). On these constructs the marker gene, xy1E is expressed from the lambda promoters pl or pr controlled by the temperature-sensitive lambda repressor cI857. A range of culture and direct detection methods were developed for tracing released hosts in lakewater (Morgan et al; 1989; 1991). As a result of using both direct detection and culture methods, the xy1E gene carried by marker plasmids was considered to be a valid indicator for detecting and studying the survival of Pseudomonas putida populations after release into sterile or untreated lake-water. (Winstanley et al., 1991). In these studies the effects of inoculum size, auxotrophic mutation carried by the host strain, metabolic burden imposed on cells by unregulated expression of xy1E, and the reproducibility of the experiments was investigated.
