The Use of H.P.L.C. for Structural Studies on Collagen

The establishment of collagen primary structure poses specific challenging problems that are linked to the size of collagen polypeptide chains, their unique amino acid compositions and repetitive sequences. The purification of the fragments needed for sequencing is particularly difficult with standard chromatographic techniques. We have thus developed a comprehensive H.P.L.C. approach for the purification of collagen chains and their CNBr and other proteolytic fragments.
