Phenotypic Changes Occurring during in vitro Activation of Human Splenic B Lymphocytes
The present study had two motivations: first to develop a system in which normal B cell differentiation could be studied in vitro. Second, to find out which antigens were the hallmarks of individual events in B cell develop-ment, with the hope that likely candidates for receptors could be identified. We show that highly purified splenocytes can be activated by anti-Ig antibody in vitro (1–2) and the phenotypic changes induced by activation defined. When T cell help was provided, these cells could be driven to differentiate (produce Ig) and a further characteristic set of phenotypic changes were shown to occur. Finally, an example is given of how this system can be used to study the function of one of the receptors (IL-2 receptor) induced by activation.
