Abstract
Approaches to the molecular cloning of DNA complementary to mRNA and chromosonal genes have advanced greatly over the past 15 yr. These advances have consisted largely of (a) the development of enzymatic methods to synthesize full length, or near full length, cDNA copies of mRNA, (b) the preparation of recombinant genomic DNA in cosmid vectors that accommodate 30–40 kilobases (kb) of genomic sequence, (c) the development of bacteriophage vectors that express the genes at the level of their encoded proteins, allowing for the identification of recombinant DNA by immunostaining expressed fusion proteins, and (d) the application of synthetic oligonucleotides complementary to predicted RNA sequences encoding known amino acid sequences for the identification of various species of recombinant DNA.
