Mechanisms of Xanthine Actions in Models of Asthma
Theophylline has long been one of the most effective and frequently used drugs for the treatment of asthma. Recently another xanthine, enprofylline, has been found useful for the treatment of asthma (Pauwels, et al, 1985). Unfortunately, the mechanism of action of xanthines in the lung remains unclear. Inhibition of cyclic nucleotide phosphodiesterase (PDE) activity was originally considered the mechanism of theophylline’s anti-asthmatic effects (Piafsky and Ogilvie, 1975). However, it has been claimed that at therapeutic plasma levels theophylline does not appreciably inhibit PDE activity and thus PDE inhibition cannot account for the clinical effects of theophylline (Persson, et al, 1986).
