Abstract

In 1963 it was shown that nalidixic acid inhibited DNA synthesis, but not until 1976 was the intracellular “target” protein identified as DNA gyrase. Until that time DNA gyrase had not even been identified as a cellular component, but subsequent studies have shown this enzyme to be an essential component of the bacterial cell and related type II DNA topoisomerases to be essential for all cellular systems.
