Dendritic Cell Vaccines for Immunotherapy of Cancer: Challenges in Clinical Trials

Abstract

Dendritic cells (DCs) are potent activators of immunity. After years of careful laboratory research and painstaking work to define cell culture parameters that allow these cells to be expanded from precursors in vitro, DCs have been tested clinically. Over the last decade, they have demonstrated a clear ability to prime and boost immunity to foreign and self antigens in vitro and in vivo. Clinical trials have also indicated that DC-based vaccines can be effective in promoting clinically relevant antitumor immunity in a small percentage of cancer patients, but these important cells must be better understood for their full clinical potential to be realized.
