Summary
Benzidine (BZD) and 4,4″-diaminoterphenyl (DAT) were examined for their mutagenicity in V79 cells (HGPRT deficiency). A very weak, but statistically significant response was obtained with DAT, a more clear effect was obtained with BZD, but only in the presence of a mouse liver S9 fraction. BZD, DAT, 4-dimethylaminoazobenzene (DAB) and 4-cyanodimethylaniline (CDA) were tested with the alkaline elution technique for damage induced in vivo in rat liver DNA. Only DAT was negative in this test. However, DAT was positive (even more than BZD) in inducing DNA damage in the rat bladder. Rat liver DNA was also tested for alkaline DNA unwinding, evaluated viscosimetrically. Only DAB, CDA and BZD were tested. A positive result was observed only for BZD.
