The Testing of four UKEMS Trial Compounds for Bacterial Mutation in the Salmonella/Microsome Assay Using (1) Aroclor-induced Rat Liver, (2) Chicken Liver S9 and (3) The York Modification of the Standard Ames Protocol

Abstract

The testing of four UKEMS trial compounds in the presence of Aroclor 1254-induced rat liver S9 using the standard Ames protocol showed that BZD and DAT induced mutation in S. typhimurium TA1538. A modification of the standard testing procedure known as the ‘York method’ demonstrated enhanced mutagenicity with BZD but not with DAT; Sample B of CDA was rendered a mutagen using this procedure. Optimum mutagenicity was seen with a final S9 concentration of 10%. Chicken liver S9 was less effective than rat in activating the two positive mutagens.
