Expression of Carp α-Globin Gene in Tilapia Ovary Cells
Abstract
For studying the expression of the carp Cyprinus arcpio α-globin gene which contains the 3′- and 5′-flanking regions, 2 introns and 3 exons [1] in cultured fish cells, the carp α-globin gene was combined with the pSVO expression vector plasmid. This recombinant DNA was transfected into TO-2 cells derived from the ovary of the tilapia hybrid Tilapia mossambica X T. nilotica [2] by DNA-calcium phosphate coprecipitation. After transfection, the transcription of the introduced carp a-globin gene in TO-2 cells was analyzed. Results of dot-blot and Northern-blot hybridization of RNA showed that the carp α-globin gene had been successfully introduced and transcribed in the cultured fish cells. This means that the expression regulatory sequence of this gene can fulfill its function in the non-erythroid fish cells.
