Detection of Pseudomonas savastanoi pv. savastanoi (Pss) on Asymptomatic Olive Plant Tissues by Enrichment-PCR

Abstract

Pseudomonas savastanoi pv. savastanoi (Pss) incites the olive knot disease characterized by tumorous outgrowths, that is dependent on bacterial production of the phytohormone indoleacetic acid (IAA) and cytokinins. Detection of Pss is currently based on bacterial isolation followed by pathogenicity tests, biochemical and/or serological identification. These conventional methods are time-consuming and expensive, requiring bacterial isolation. We have used the published sequence of iaaL gene to design specific primers for amplification of this gene, and developed a sensitive and specific detection method for Pss with a previous bacterial enrichment step (Enrichment-Standard PCR). We have also set up a nested-PCR assay in a single closed tube (Nested-PCR) for detection of Pss.
