Structure-Activity Relationships in Doxorubicin Related Compounds

Abstract

Doxorubicin (Ia) belongs to the anthracycline family of antibiotics, not rarely encountered as pigmented products of the metabolism in cultures of different species of the genus Streptomyces. The classical anthracyclines such as the rhodomycins, the pyrromycins and the cinerubins, although displaying cytotoxic activity, are not clinically useful compounds because of their low therapeutic index. On the other hand, daunorubicin (Ib) and especially doxorubicin have received considerable attention because of their efficacy in the chemotherapic treatment of different human cancers. The use of doxorubicin in the clinical practice is however not devoid of undesirable side effects (1).
