Viral Diagnosis by Electron Microscopy

The electron microscope (EM) technique of negative staining has become an increasingly useful tool in the viral diagnostic field not only for reaching a primary diagnosis but also for corroborating the findings produced by more recent techniques. In carrying out negative staining attention to detail is important and this is discussed. The areas where negative staining can be most meaningfully employed are considered and three are delineated. These are, one, where no test based on serology exists, two, where the specimen has an unknown aetiology, and finally, in order to establish a diagnosis with problem specimens. Examples of each of these categories are given and illustrations provided.
