Integration of HGH Gene in Transgenic Mice and Transmission to Next Generation

Summary

Transgenic mice carrying the mouse metallothionein-I promoter (mMT-I)/human growth hormone (hGH) fusion gene (MThGH) were produced by microinjection into the pronucleus of fertilized one-cell eggs.

A total of eight transgenic mice (28 %) were obtained which all contained detectable MThGH-gene construct sequences in their genome, ranging from 1 or less (mosaics) to 4 copies/cell. One male transgenic mouse exhibiting high, metal-inducible levels of human growth hormone in serum and growing twice as large as control litter-mates was bred to three normal females. Eleven of twenty offspring reared were shown to be transgenic by Southern- and dot blot analyses. They had heritated the MThGH-gene construct from their father.
