The chemistry of azolotetrazinones

Abstract

In 1978, it was believed1,2that the 1,2,3,5-tetrazine ring was inherently unstable, despite claims3 of the preparation of this system.Indeed, there is only one confident report4of a [2,5-dihydro]-l,2,3,5-tetrazine without fusion to an azole. In 1979, Ege5published the first report of pyrazolo[5,1-d]-l,2,3,5-tetrazin-4(3H)-ones. There followed comprehensive reports6–8 by the Aston group of the synthesis and antitumour activity of 3-alkyl and 3-aryl-8-substitutedimidazo[5,1-d]-1,2,3,5-tetrazin-4(3H)-ones and an indazolotetrazinone.
