The Effect of DNA Methylation on DNA-Protein Interactions and on the Regulation of Gene Expression
Abstract
The transcriptional activity of a gene can be controlled in a number of ways and modulated by different factors. The interaction of specific proteins with highly specific sequence elements (“motifs”) in the promoter regions of genes plays a pivotal role in this regulation (see Schlokat and Gruss, this Vol., and Beato, this Vol.). In addition, the methylation of promoter sequences can cause the inactivation of genes in a number of eukaryotic systems (for reviews see Razin and Riggs 1980; Doerfler 1981, 1983; Riggs and Jones 1983). This chapter discusses the role of site-specific promoter methylations in the control of gene expression. (For detailed information on the biochemistry and biology of DNA methylation, see Taylor 1984; Adams and Burdon 1985; Trautner 1984; Razin et al. 1984; and Cantoni and Razin 1985).
